To validate the limit of detection of the detection of low-frequency variants, we constructed for influenza A(H1N1)pdm09 and A(H3N2) reverse genetics samples a wild type variant and a mutant variant that included known antiviral resistance mutations, namely H275Y and E119V. These wild types and mutants are used in different proportions to identify a threshold.

为验证低频变异检测的检出限(limit of detection),我们针对甲型H1N1 pdm09流感病毒与甲型H3N2流感病毒的反向遗传学(reverse genetics)样本,构建了分别携带已知抗病毒耐药突变H275Y和E119V的野生型变异株与突变型变异株。随后将这些野生型与突变型样本按不同比例混合,以确定检测阈值。
