ATAC-seq of 8-cell embryo samples

We hypothesized that LincGET/CARM1 complex activates ICM-specific genes at the 8-cell stage. And we examined chromatin accessibility of these genes by preforming an assay for transposase-accessible chromatin with high-throughput sequencing (ATAC-seq) Overall design: The LincGET- or Carm1-overexpressing blastomeres as well as control blastomeres with EGFP overexpressing at the 8-cell stage were collected and peformed for ATAC-seq.我们提出假说：LincGET/CARM1复合物可在8细胞阶段激活内细胞团（Inner Cell Mass，ICM）特异性基因。我们通过转座酶可及性染色质高通量测序（Assay for Transposase-Accessible Chromatin with high-throughput Sequencing，ATAC-seq）检测了上述基因的染色质可及性。整体实验设计：收集过表达LincGET或Carm1的卵裂球，以及过表达EGFP的对照卵裂球，所有样本均采集于8细胞阶段，并进行ATAC-seq检测。
