We aim to further elucidate the effect of succinate and the succinate dehydrogenase inhibtor, dimethyl malonate, on macrophages treated with LPS. Specifically we want to deepen our understanding of how these reagents influence both LPS-induced IL-1B and IL-10. Overall design: BMDMs were either left untreated or treated with succinate or dimethyl malonate for 3 h prior to treatment with or without LPS for 4 or 48 h.
本研究旨在进一步阐明琥珀酸(succinate)与琥珀酸脱氢酶抑制剂丙二酸二甲酯(dimethyl malonate)对脂多糖(Lipopolysaccharide, LPS)刺激的巨噬细胞的作用效果。具体而言,本研究拟深化对这两种试剂如何调控脂多糖诱导的白细胞介素1β(Interleukin-1β, IL-1β)与白细胞介素10(Interleukin-10, IL-10)表达的认知。实验设计:将骨髓来源的巨噬细胞(Bone Marrow-derived Macrophages, BMDMs)分为未处理对照组与实验组,分别经琥珀酸或丙二酸二甲酯预处理3小时后,再以脂多糖处理或不予处理,继续培养4小时或48小时。
