Elevated intracellular cyclic-di-GMP level in Shewanella oneidensis increases expression of c-type cytochromes

Transcriptomic analysis used to understand the influence of elevated intracellular cyclic-di-GMP in Shewanella oneidensis on expression of c-type cytochrome at transcript level. RNA were extracted from S. oneidensis MR-1 WT and pYedQ2 cultures using the RNeasy mini Kit (Qiagen, Germany), with the DNA being depleted using RNase-free DNase set (Qiagen, Germany), sequenced using Illumina HiSeq2500.

本研究采用转录组分析技术，解析奥奈达希瓦氏菌（Shewanella oneidensis）中细胞内环二GMP（cyclic-di-GMP）水平升高对c型细胞色素（c-type cytochrome）转录水平表达的影响。实验以奥奈达希瓦氏菌MR-1野生型（WT）和pYedQ2菌株的培养物为材料，使用德国Qiagen公司的RNeasy 迷你试剂盒（RNeasy mini Kit）提取RNA，通过无RNase脱氧核糖核酸酶试剂盒（RNase-free DNase set，德国Qiagen公司）去除基因组DNA，随后利用Illumina HiSeq2500测序平台完成测序。
