Fecal samples from 35 acromegaly patients and 25 control patients (with sellar region lesions but no pituitary dysfunction) were subjected to 16S rRNA sequencing and metabolomics analysis. Subsequently, fecal microbiota from 20 randomly selected human donors (10 per group) was transplanted into 20 germ-free mice, followed by single-cell sequencing analysis of colonic tissues. The fecal samples from these 10 acromegaly patients and 10 control subjects were then analyzed using metagenomics and metaproteomics. Additionally, intestinal polyp samples from 6 acromegaly patients and 6 control patients were selected for proteomic analysis. Finally, target bacteria were transplanted into germ-free mice, and single-cell sequencing analysis was performed on the colonic tissues of these mice.
本研究收集了35例肢端肥大症(acromegaly)患者与25例对照患者(存在鞍区病变但无垂体功能障碍)的粪便样本,对其开展16S rRNA测序(16S rRNA sequencing)与代谢组学分析。随后,随机选取20名人类供体的粪便菌群(每组各10例)移植至20只无菌小鼠(germ-free mice)体内,继而对小鼠的结肠组织进行单细胞测序分析。此后,针对上述10例肢端肥大症患者与10例对照受试者的粪便样本,采用宏基因组学(metagenomics)与宏蛋白质组学(metaproteomics)进行分析。此外,选取6例肢端肥大症患者与6例对照患者的肠道息肉样本实施蛋白质组学分析。最终,将目标细菌移植至无菌小鼠体内,并对该类小鼠的结肠组织开展单细胞测序分析。
