Transcriptional profile of PCSC spheres in SCM-1% KO (stem-like cells) vs adherent cultures in PCSC-Celprogen medium (differentiated-like cells) Two-condition experiment: Sphere vs. Parental/adherent cells. Biological replicates: 2 sphere replicates , 2 adherent replicates. Overall design: Early passaged tumor cells were expanded in adherent conditions and then plated in sphere forming conditions (SCM-1KO% medium at low density) to isolate sphere growing cells.
本数据集为两组样本的转录组谱对比实验:一组为SCM-1%基因敲除(SCM-1% KO)环境中培养的PCSC球状体(干细胞样细胞,stem-like cells),另一组为PCSC-Celprogen培养基(PCSC-Celprogen medium)中贴壁培养的分化样细胞(differentiated-like cells)。本实验为双条件对照实验,即球状体细胞与亲本/贴壁细胞的对比。生物学重复设置:球状体细胞组与贴壁细胞组各设2次生物学重复。实验整体设计:将低代次肿瘤细胞先在贴壁培养条件下扩增,随后以低密度接种于SCM-1%基因敲除培养基中进行球状体形成培养,以分离获得球状体增殖细胞。
