To construct the optimal fatty liver cell model in vitro and use TMAO intervention to observe its effect on fatty liver cells and identify potential key genes for change by transcriptomics. Overall design: Comparative gene expression profiling of RNA-seq data between HepG2 fatty liver cells and HepG2 fatty liver cells with TMAO intervention, three samples in each group.
本研究旨在构建最优的体外脂肪肝细胞模型,通过三甲胺N-氧化物(Trimethylamine N-oxide,TMAO)干预观察其对脂肪肝细胞的影响,并借助转录组学(transcriptomics)技术,利用RNA测序(RNA-seq)数据筛选鉴定潜在的差异表达关键基因。实验设计概述:针对两组样本的RNA测序数据开展基因表达谱对比分析,两组分别为HepG2脂肪肝细胞组,以及经TMAO干预的HepG2脂肪肝细胞组,每组设置3个生物学重复样本。
