Chromatin function of IkBa: ChIP-on-chip of IkBa

The Inhibitor of the NFkB, IkBa, is the main regulator of the NFkB pathway. Our data has demonstrated that there is an alternative function for IkBa in the chromatin that affects HOX and other developmental genes. Keywords: CHIP-chip Chip-on-chip analysis has demonstrated that IkBa associates to the promoters of several genes. Chromatin fragments were obtained from NIH-3T3 cells and precipitated with 3 different IkBa comercial antibodies (raised in mouse, goat and rabbit), and fold enrichment was compared to input.

核因子κB（NFkB）抑制剂IκBα（IkBa）是NFκB通路的主要调控因子。本研究数据证实，IκBα在染色质中存在非经典功能，可调控同源框基因（HOX）及其他发育相关基因。
关键词：染色质免疫沉淀-芯片（ChIP-chip，Chip-on-chip）分析证实，IκBα可结合多个基因的启动子区域。实验中从NIH-3T3细胞系中获取染色质片段，使用3种商品化IκBα抗体（分别由小鼠、山羊和兔源免疫制备）进行免疫沉淀，并将富集倍数与输入对照样本进行对比。
