The differential expression of the high-yield astaxanthin Phaffia rhodozyma BPAX-A1 with the wild-type Phaffia rhodozyma CBS6938 was obtained and analysed to understand metabolic changes leading to an improved astaxanthin production. Overall design: We performed fermentations in instrumented fermenters with wild-type and mutant strains (three biological replicates with each strain) and samples were obtained at the late exponential growth phase for RNA-seq analysis- this phase presented the highest astaxanthin production rate suggesting to be the most informative to explain metabolic changes leading to an improved astaxanthin production in the mutant strain.

本研究对高产虾青素的红发夫酵母(Phaffia rhodozyma)BPAX-A1与野生型红发夫酵母CBS6938的基因差异表达情况进行了检测与分析,以阐明介导虾青素合成能力提升的代谢变化。 实验设计:本研究在受控发酵罐中开展野生型与突变型菌株的发酵培养,每个菌株设置3次生物学重复;于指数生长后期采集样本进行RNA测序(RNA-seq)分析。该时期菌株的虾青素合成速率最高,是解析突变株虾青素产量提升相关代谢变化的最优采样阶段。
