Pseudotsuga menziesii parental trees used in this study were obtained from provenances originating from USA Washington, either from North Bend (trees 4440, 4455 and 4456) or Enumclaw (trees 4466, 4474 and 4477). The following control crosses were performed at INRA, Orléans, France: 4474 x 4440, 4455 x 4466, and 4456 x 4477. Somatic embryogenesis induction from immature zygotic embryos was performed according to Reeves et al (2018), in 2011 to obtain the SD4 primary (1ry) embryogenic line (4456 x 4477) and in 2012 to obtain 1ry embryogenic lines TD15 (4474 x 4440) and TD17 (4455 x 4466). The 1ry lines SD4, TD15 and TD17 generated secondary (2ry) EM and NE lines respectively labelled SD4-8EM and SD4-8NE, TD15-1EM and TD15-1NE, TD17-1EM and TD17-1NE.
本研究所用的花旗松(Pseudotsuga menziesii)亲本树木采自美国华盛顿州的两个种源地,分别为北本德(对应树木编号4440、4455与4456)和埃努姆克拉夫(对应树木编号4466、4474与4477)。本研究的对照杂交实验于法国奥尔良的法国国家农业食品与环境研究院(INRA)开展,共设置3个杂交组合:4474 × 4440、4455 × 4466及4456 × 4477。参照Reeves等(2018)建立的方法,本研究通过未成熟合子胚诱导体细胞胚胎发生:2011年获得杂交组合4456 × 4477的初级(1ry)胚性株系SD4;2012年分别获得杂交组合4474 × 4440的初级胚性株系TD15,以及杂交组合4455 × 4466的初级胚性株系TD17。上述初级胚性株系SD4、TD15与TD17分别产生了次级(2ry)胚性块(EM)与非胚性块(NE)株系,对应的命名依次为SD4-8EM与SD4-8NE、TD15-1EM与TD15-1NE、TD17-1EM与TD17-1NE。
