Prolonged cold exposure stimulates the recruitment of beige adipocytes within white adipose tissue. Beige adipocytes depend on mitochondrial oxidative phosphorylation to drive thermogenesis. The transcriptional coregulator TLE3 inhibits mitochondrial and metabolic gene expression in beige adipocytes. mRNA profiles of iWAT immortalized preadipocytes, differentiated in culture, and knocking out TLE3 after differentiation, were generated by deep sequencing, in triplicate, using Illumina HiSeq 2500.
长期冷暴露可诱导白色脂肪组织内米色脂肪细胞(beige adipocytes)的募集。米色脂肪细胞依赖线粒体氧化磷酸化完成产热功能。转录共调控因子TLE3能够抑制米色脂肪细胞中线粒体及代谢相关基因的表达。本研究以体外分化得到的腹股沟白色脂肪组织(inguinal white adipose tissue, iWAT)永生化前体脂肪细胞为研究对象,在分化完成后对其进行TLE3敲除处理,随后采用Illumina HiSeq 2500平台开展三次生物学重复的深度测序,由此获取上述样本的mRNA表达谱。
