To investigate biofilm-leukocyte crosstalk, we performed RNA-seq to evaluate how the S. aureus biofilm transcriptome was altered following exposure to G-MDSCs or macrophages in vitro. Interestingly, each immune cell type elicited a distinct transcriptional response. Overall design: RNA-sequencing was performed on biofilm co-cultured with G-MDSCs (3 replicates), macrophages (3 replicates), or G-MDSCs + macrophages (2 replicates). Biofilm only was also used as a control (2 replicates).
为探究生物膜与白细胞的串扰(biofilm-leukocyte crosstalk),我们开展了RNA测序(RNA-seq)实验,以评估体外暴露于粒细胞-髓系来源抑制细胞(granulocytic myeloid-derived suppressor cells, G-MDSCs)或巨噬细胞后,金黄色葡萄球菌(Staphylococcus aureus, S. aureus)生物膜的转录组变化。值得注意的是,两种免疫细胞类型分别诱导了截然不同的转录应答。实验设计概述:我们对以下样本进行了RNA测序:与G-MDSCs共培养的生物膜(3个生物学重复)、与巨噬细胞共培养的生物膜(3个生物学重复)、与G-MDSCs联合巨噬细胞共培养的生物膜(2个生物学重复),以及仅培养的生物膜对照组(2个生物学重复)。
