DNA binding activities of the C-clamp

Investigated the DNA binding specificity of the C-clamp of dnTCF1E Used a doxycycline inducible system to investigate the role of the C-clamp in the genome-wide binding of dnTCF1E. Induced dnTCF1EWT and dnTCF1Emut (C-clamp mutant that is null for C-clamp DNA binding) over 2 time points (2 hours and 9 hours). A 0 hour (no doxycycline, uninduced) control was also included.

本研究针对dnTCF1E的C-clamp结构域的DNA结合特异性展开探究，并采用强力霉素（doxycycline）诱导表达系统，解析C-clamp结构域在dnTCF1E全基因组结合过程中的功能。实验分别在2小时、9小时两个时间点诱导野生型dnTCF1E（dnTCF1EWT）与突变型dnTCF1E（dnTCF1Emut，其C-clamp完全丧失DNA结合活性）的表达，同时设置0小时（未添加强力霉素、未诱导）的对照组。
