Chronically-instrumented conscious Wistar rats: 6 female Wistar rats. Dofetilide per os (3 and 5 mg/kg). No PK performed. Haemodynamic parameters (SAP, DAP, MAP, HR), lead II ECG (RR, PR, QRS, QT, QTcB and QTcF intervals) and body temperature + arrhythmias. Arterial blood pressure, heart rate, body temperature and ECG parameters were measured 45, 30 and 15 min before administration (mean of the measurements obtained at these time points = baseline; T0) and 30 (T30), 60 (T60), 120 (T120), 180 (T180), 240 (T240), 360 (T360), 600 (T600) and 1440 min (T1440) post-administration.
经慢性植入手术的清醒Wistar大鼠:实验动物为6只雌性Wistar大鼠。受试药物为多非利特(Dofetilide),采用口服给药方式,设置剂量为3 mg/kg与5 mg/kg。本实验未开展药代动力学(Pharmacokinetics, PK)相关检测。实验观测指标包括血流动力学参数:收缩压(SAP, Systolic Arterial Pressure)、舒张压(DAP, Diastolic Arterial Pressure)、平均动脉压(MAP, Mean Arterial Pressure)、心率(HR, Heart Rate);Ⅱ导联心电图(ECG)参数:RR间期、PR间期、QRS波时限、QT间期、巴泽特校正QT间期(QTcB)、弗雷明汉校正QT间期(QTcF),同时记录大鼠体温与心律失常发生情况。动脉血压、心率、体温及心电图参数分别于给药前45 min、30 min、15 min时采集,取上述三个时间点的测量均值作为基线值(记为T0);给药后分别于30 min(T30)、60 min(T60)、120 min(T120)、180 min(T180)、240 min(T240)、360 min(T360)、600 min(T600)及1440 min(T1440)进行数据采集。
