Seasonal Dynamics of Hedgehog-Borne Ticks and Severe Fever with Thrombocytopenia Syndrome Virus in Beijing’s Urban Parks

All data were collected from H. longicornis ticks that were sourced from Beijing in 2024. For viral data collection, total RNAs were extracted from tick homogenates and hedgehog blood cells using either TRIzol reagent (Thermo Fisher Scientific, USA) or the RNeasy kit (Qiagen, Germany), in accordance with the respective manufacturer's protocols. The RNA samples were then reverse transcribed using a One-Step SYBR PrimerScript reverse transcription (RT)-PCR kit (TaKaRa, Japan). Following reverse transcription, the target viral fragments were amplified using the nested PCR method. The resulting PCR products were then sequenced by the Sanger method for further analysis.

本研究所有数据均采集自2024年采自北京的长角血蜱（Haemaphysalis longicornis）。在病毒数据采集环节，研究人员依照对应厂商的操作规范，使用TRIzol试剂（赛默飞世尔科技，美国）或RNeasy试剂盒（凯杰，德国）从蜱组织匀浆及刺猬血细胞中提取总RNA。随后，采用一步法SYBR PrimeScript反转录聚合酶链式反应（RT-PCR）试剂盒（宝生物工程，日本）对RNA样本进行反转录。反转录完成后，通过巢式PCR法扩增目标病毒片段。所得PCR产物随后采用桑格测序法进行测序，以开展后续分析。
