brain tissue from all subregions (CA1, CA3, dentate gyrus) of the hippocampal formation was isolated at postnatal day 8 and at an adult age (3-6months old animals) in wildtype and NEIL3-deficient mice (n=4 at p8 and n=3 at adult age). NEILs are DNA glycosylases potentially involved in transcription regulation and hippocampal neuronal maturation. RNA was extracted and sequenced. Wildtype (C57Bl6) and mice deficient in NEIL3 (C57Bl6-background) were used. Tissue was isolated exclusively at baseline, i.e. the animal was taken directly from its homecage without any behavioral intervention
本研究分别于出生后第8天与成年阶段(3~6月龄动物),采集野生型及NEIL3缺陷型小鼠的海马结构(hippocampal formation)全部亚区——CA1、CA3及齿状回(dentate gyrus)的脑组织;其中出生后第8天组样本量为4只,成年组为3只。NEIL家族蛋白属于DNA糖苷酶(DNA glycosylases),可能参与转录调控及海马神经元成熟过程。实验对提取的RNA进行测序。所用野生型小鼠为C57BL/6品系,NEIL3缺陷型小鼠均为C57BL/6背景。组织仅在基线状态下取材,即直接从动物饲养笼中取样,未施加任何行为学干预。
