Our goal was to determine whether osteoblastic LuCaP 23.1 prostate cancer xenograft tumors can elicit an osteoblastic response at the gene expression level in human bone marrow stromal cells. Custom Agilent 44K whole human genome expression oligonucleotide microarrays were used to profile bone marrow stroma cells isolated from three patients and treated with either mineralization media or LuCaP 23.1 conditioned medium. Total RNA was isolated and amplified prior to hybridization against a common reference pool of prostate tumor cell lines.
本研究旨在探究成骨细胞型LuCaP 23.1前列腺癌异种移植瘤能否在人类骨髓基质细胞中于基因表达层面诱导成骨细胞应答反应。本研究采用定制安捷伦(Agilent)44K全人类基因组表达寡核苷酸微阵列,对3名患者分离得到的骨髓基质细胞开展表达谱分析,这些细胞分别经矿化培养基或LuCaP 23.1条件培养基处理。在与前列腺肿瘤细胞系通用参考混合池进行杂交前,已完成总RNA的分离与扩增操作。
