This study identifies progression in breast ductal carcinoma in situ (DCIS) as it progresses towards triple negative invasive breast cancer (TNBC). Overall design: Single cell RNA sequencing (scRNAseq) and bulk RNA sequencing was performed on the C3Tag genetically engineered mouse model that forms human breast-like DCIS and TNBC. Bulk RNA sequencing was performed on RNA harvested from 10 archival formalin-fixed paraffin embedded (FFPE) pairs comprising of estrogen receptor (ER) positive (ER+) and ER- subsets of DCIS.

本研究聚焦乳腺导管原位癌(breast ductal carcinoma in situ, DCIS)向三阴性浸润性乳腺癌(triple negative invasive breast cancer, TNBC)的进展过程。实验总体设计如下:采用可构建类人乳腺DCIS与TNBC模型的C3Tag基因工程小鼠,开展单细胞RNA测序(single cell RNA sequencing, scRNAseq)与批量RNA测序(bulk RNA sequencing);同时对10对存档福尔马林固定石蜡包埋(archival formalin-fixed paraffin embedded, FFPE)样本进行研究,该批样本涵盖雌激素受体阳性(ER+)与雌激素受体阴性(ER-)两类DCIS亚组,对其提取的RNA开展批量RNA测序。
