The goal of this study is to compare gene expression of mouse cultured cortical neurons following exposure to extracellular miR-124-5p, and miR-92a-1-5p, which are dysregulated in Alzheimer´s disease. Overall design: To investigate the effect of extracellular miRNAs on neurons, we exposed mouse cortical neurons to miR-92a-1-5p, miR-124-5p and a mutated control oligonucleotide, Mut.oligo, for 6h and 5d. Total RNA was extracted for RNA-seq, in 4 replicates, and comparative gene expression profiles were analyzed.

本研究旨在比较暴露于细胞外微小RNA-124-5p(miR-124-5p)与微小RNA-92a-1-5p(miR-92a-1-5p)后的小鼠培养皮层神经元的基因表达水平,这两种微小RNA在阿尔茨海默病中存在表达失调情况。 实验设计概述:为探究细胞外微小RNA对神经元的影响,我们将小鼠皮层神经元分别暴露于miR-92a-1-5p、miR-124-5p及突变型对照寡核苷酸Mut.oligo中,处理时长为6小时与5天。随后提取总RNA进行RNA测序(RNA-seq),实验设置4次重复,并对比较基因表达谱展开分析。
