Obesity is associated with various lung pathologies. We performed single nucleus RNA sequencing of formalin-fixed lungs of mice to obtain insights on this. Overall design: Groups of male and female mice of about 6 months age had been kept on ad libitum for about 3 months on high-fat or control diet (Bioserv products S3282 and F4031, with 60% and 10% fat calories respectively), when they were sacrificed with carbon dioxide-based euthanasia. Lungs were immediately dissected, rinsed in PBS, and the left lower lung lobes flash-frozen in liquid nitrogen. Nuclei isolated from frozen tissues were used for sequencing.

肥胖与多种肺部病理状态存在显著关联。为阐明二者间的潜在关联机制,我们针对小鼠福尔马林固定肺部组织完成了单细胞核RNA测序(single nucleus RNA sequencing)实验。 实验设计概况:将约6月龄的雌雄小鼠分为两组,分别以高脂饲料与对照饲料自由采食(ad libitum)饲养约3个月,两种饲料均购自Bioserv,产品编号分别为S3282与F4031,脂肪供能占比分别为60%与10%;随后采用二氧化碳安乐死法处死小鼠。解剖获取肺部组织后,立即用磷酸盐缓冲液(Phosphate Buffered Saline, PBS)漂洗,将左肺下叶置于液氮中快速冷冻。从上述冷冻组织中分离得到的细胞核将用于后续测序实验。
