In this study we aimed to determine denitrifier and DNRA community structures in contrasting German soils by 16S rRNA and functional gene (narG, napA, nirK, nirS, nosZI, nosZII and nrfA) amplicon sequencing, quantify those gene via qPCR, assess apparent Michaelis-Menten kinetics for denitrification and N2O reduction and determine key taxa and indicator genes for denitrification and N2O reduction. Relating microbial community composition to reaction kinetics can help to reveal important ecological drivers of microbial communities and their activities, and will provide valuable data for denitrification models.
本研究旨在通过16S核糖体RNA(16S rRNA)与功能基因(narG、napA、nirK、nirS、nosZI、nosZII及nrfA)扩增子测序,解析德国不同类型土壤中的反硝化微生物与异化硝酸盐还原为铵(DNRA)群落结构;同时采用实时定量聚合酶链反应(quantitative PCR,qPCR)对上述基因开展定量分析,评估反硝化与一氧化二氮(N₂O)还原过程的表观米氏动力学参数,并明确反硝化及N₂O还原过程的关键类群与指示基因。将微生物群落组成与反应动力学相关联,有助于揭示调控微生物群落及其功能活动的核心生态驱动因子,同时可为反硝化模型构建提供极具价值的参考数据。
