We investigated how an intron with AT-AC terminal sequences is spliced using th Arabidopsis drol1 mutant and its suppressor sudl1-1. The terminal sequences of the AT-AC intron were replaced with GT-AG, GT-AC, and AT-AG and the splicing patterns were examined by PCR and sequencing. We also investigated the splicing properties of the suppressor using a recombinant gene with duplicated intron termini.
本研究以拟南芥drol1突变体及其抑制突变体sudl1-1为实验材料,探究了带有AT-AC末端序列的内含子的剪接机制。我们将AT-AC内含子(AT-AC intron)的末端序列分别替换为GT-AG、GT-AC与AT-AG,并通过聚合酶链式反应(PCR)与测序技术分析其剪接模式。此外,我们还借助带有重复内含子末端的重组基因,探究了该抑制突变体的剪接特性。
