Oxygen consumption rates were measured in L6E9 myoblasts and R1H cells culture in growth medium or in differentiation medium at 37°C by polarography in DMEM serum free (basal), in the presence of oligomycin, and after the addition of FCCP. Respiration rates are expressed in nmol O2.min−1.cell−1. Respiratory control ratio (RCR) was calculated as the ratio between FCCP-stimulated and oligomycin insensitive respiration rates. For more details see Materials and Methods. Data are means ± SE from 12 independent culture dishes. *P<0.05, significantly different from corresponding cells in GM. † P<0.05, †† P<0.01 and ††† P<0.001: significantly different from L6E9 cells.

本研究采用极谱法,于37℃条件下,对生长培养基(GM)及分化培养基中培养的L6E9成肌细胞与R1H细胞的耗氧速率进行检测,检测体系分别为无血清DMEM(基础状态)、添加寡霉素时,以及加入羰基氰化物间氯苯腙(FCCP)后。呼吸速率以nmol O₂·min⁻¹·cell⁻¹为单位进行表示。呼吸控制比(RCR)通过FCCP刺激后的呼吸速率与寡霉素不敏感呼吸速率的比值计算得到。详细实验步骤参见材料与方法部分。实验数据为12个独立培养皿样本的均值±标准误(SE)。*P<0.05表示与生长培养基(GM)中对应的细胞存在显著性差异;†P<0.05、††P<0.01及†††P<0.001表示与L6E9细胞存在显著性差异。
