In axolotls, stem cells are systemically prompted to re-enter the cell cycle upon amputation. We sought to identify how the cells surrounding the systemically activated cells respond to an amputation. Overall design: We dissociated homeostatic or systemically-activated limbs at 14 days post-amputation, and sorted them by their DNA content using FACS. We then performed single-cell RNA sequencing of the dividing, 2C population using 10X genomics.
