Spatial transcriptomics was performed in areas enriched with CD45+ immune cells or CD8+ T cells. Data was generated with 12 mice: 4 NSG-SGM3 mice reconstituted with blood fo 4 healthy individuals, 4 NSG-SGM3 mice reconstituted with blood from 4 PsA patients and 4 NSG-SGM3 mice reconstituted with blod cells of 4 Ps patients. Overall design: In the study we enrolled 4 healthy individual, 4 patients with psoriasis and 4 patients with Psoriatic Arthritis . Skin lesions and artritis were evaluated with the Psoriasis Area Severity Index (PASI) Score and the extent of PsA with the Disease Assessment in Psoriatic Arthritis (DAPSA) score. We identified 1813 genes using the GeoMx platform in mouse synovail tissue collected from NSG-SGM3 mice engrafted with human PBMC and sera from healthy individuals, psoriais and Psoriatic arthritis patients.
本研究针对富集CD45阳性免疫细胞(CD45+ immune cells)或CD8阳性T细胞(CD8+ T cells)的区域开展空间转录组学(Spatial transcriptomics)检测。本研究以12只NSG-SGM3小鼠为实验对象生成测序数据:其中4只通过输注4名健康个体的外周血完成免疫重建,4只通过输注4名银屑病关节炎(Psoriatic Arthritis, PsA)患者的外周血完成免疫重建,剩余4只通过输注4名银屑病(Psoriasis, Ps)患者的血细胞完成免疫重建。总体实验设计:本研究共纳入4名健康个体、4名银屑病患者及4名银屑病关节炎患者。皮肤病变采用银屑病面积与严重性指数(Psoriasis Area Severity Index, PASI)评分评估,银屑病关节炎的疾病范围采用银屑病关节炎疾病评估量表(Disease Assessment in Psoriatic Arthritis, DAPSA)评分评估。本研究依托GeoMx平台,在植入人外周血单个核细胞(peripheral blood mononuclear cell, PBMC)并输注健康个体、银屑病患者及银屑病关节炎患者血清的NSG-SGM3小鼠所采集的滑膜组织中,共鉴定出1813个基因。
