Immunoglobulin secretion influences the composition of chicken caecal microbiota

In this study we tested interactions between chicken immunoglobulins and caecal microbiota in two different experiments. First we determined which chicken caecal microbiota was coated by immunoglobulins and next, using Ig deficient chickens, we identified taxa, colonisation of which was dependent on immunoglobulin expression.Using cell sorting followed by sequencing of V3/V4 variable region of 16S rRNA, bacterial species with increased or decreased immunoglobulin coating were determined. Next we determined caecal microbiota composition in immunoglobulin knockout chickens. We found out that immunoglobulin coating was common and major taxa were coated with immunoglobulin.
本研究通过两组独立实验,探究鸡免疫球蛋白(immunoglobulin)与盲肠微生物群(caecal microbiota)之间的相互作用。首先,本研究明确了鸡盲肠微生物群中哪些类群被免疫球蛋白包被;随后,利用免疫球蛋白缺陷型鸡只,鉴定出定植状态依赖于免疫球蛋白表达的微生物分类单元(taxa)。通过细胞分选后对16S核糖体RNA(16S rRNA)的V3/V4可变区进行测序,本研究确定了免疫球蛋白包被水平升高或降低的细菌物种。接下来,本研究检测了免疫球蛋白敲除鸡只的盲肠微生物群组成。研究结果显示,免疫球蛋白包被现象普遍存在,且主要微生物分类单元均会被免疫球蛋白包被。
