Confocal and micro-CT data of Angucharcotes thayerae, holotype, NIGP180055

This dataset contains the raw confocal laser scanning microscopy (CLSM) slices and X-ray microtomography (micro-CT) slices for the holotype of <em>Angucharcotes thayerae</em> (NIGP180055) from mid-Cretaceous Burmese amber. This dataset is associated with the publication "Earliest fossil record of Phloeocharinae in mid-Cretaceous amber from northern Myanmar (Coleoptera: Staphylinidae)" (DOI:10.1016/j.cretres.2022.105371). Confocal images were obtained with a Zeiss LSM710 confocal laser scanning microscope, using the 488 nm (Argon) laser excitation line. The original CZI files are provided, which could be opened by the ZEISS ZEN software. Micro-CT data were obtained with a Zeiss Xradia 520 Versa 3D X-ray microscope. Scanning parameters were as follows: isotropic voxel size, 1.4777 μm; power, 3 W; acceleration voltage, 40 kV; exposure time, 5 s; projections, 2801. The TIFF stack is provided.
本数据集包含产自白垩纪中期缅甸琥珀的*Angucharcotes thayerae*(标本编号NIGP180055,正模标本)的原始共聚焦激光扫描显微镜(confocal laser scanning microscopy, CLSM)切片与X射线显微断层扫描(X-ray microtomography, micro-CT)切片。本数据集关联学术论文《缅甸北部白垩纪中期琥珀中苔甲亚科(Phloeocharinae)最早化石记录(鞘翅目:隐翅虫科)》(DOI:10.1016/j.cretres.2022.105371)。共聚焦成像采用蔡司LSM710共聚焦激光扫描显微镜,以488 nm(氩离子)激光激发光路完成采集;原始CZI格式文件已提供,可通过ZEISS ZEN软件直接打开。显微CT数据通过蔡司Xradia 520 Versa 3D X射线显微镜采集,扫描参数如下:各向同性体素尺寸1.4777 μm,扫描功率3 W,加速电压40 kV,曝光时长5 s,投影总数2801;数据集同时提供TIFF图像序列文件。
