We have investigated the human cytomegalovirus lytic transcriptome using the Pacific Biosciences Sequel (PacBio) and the Oxford Nanopore Technologies (ONT) MinION (R9.4) platform. The sample sequenced on the PacBio platform was prepared following the IsoSeq protocol. The samples sequenced on the MinION platform include a cDNA sample that was cap-selected and a cDNA sample that was not. One RNA sample was also sequenced by the MinION sequencer in its native RNA form using direct RNA sequencing.
本研究针对人类巨细胞病毒裂解期转录组,采用太平洋生物科学公司Sequel(Pacific Biosciences Sequel,简称PacBio)与牛津纳米孔科技公司MinION(Oxford Nanopore Technologies MinION,型号R9.4)测序平台展开了分析。基于PacBio平台的测序样本依照IsoSeq实验流程制备完成。采用MinION平台测序的样本包含两份互补脱氧核糖核酸(cDNA)样本:一份为经加帽筛选的cDNA样本,另一份为未经过加帽筛选的cDNA样本。另有一份RNA样本以天然RNA形态,通过MinION测序仪完成了直接RNA测序(direct RNA sequencing)。
