We report the effect of H19 knockdown on genome wide methylation by Enhanced Reduced Representation Bisulfite Sequencing (eRBBS) to detect quantitative base pair changes. Differential methylation genome wide analysis was done on NEPC derived organoid expressing shH19 or a control vector. Overall design: Three replicates of organoids each expressing shH19 and and control vectors were utilized for ERBBS assay for differential methylation.
本研究采用增强型简化代表性亚硫酸氢盐测序(Enhanced Reduced Representation Bisulfite Sequencing,eRBBS)检测定量碱基对变化,以探究H19基因敲低对全基因组甲基化的影响。本研究针对表达shH19或对照载体的神经内分泌前列腺癌(Neuroendocrine Prostate Cancer,NEPC)来源类器官开展全基因组差异甲基化分析。实验设计:本研究共设置3组生物学重复的类器官,分别表达shH19与对照载体,用于eRBBS实验以分析差异甲基化水平。
