An uncrewed surface vessel, USV Surveyor, and an automated eDNA sample collection device, the Environmental Sample Processor, were used to obtain eDNA samples autonomously over a 4,200 km, 29 day voyage in the eastern North Pacific Ocean. We used DNA metabarcoding (Illumina MiSeq) of cytochrome c oxidase subunit 1 (COI) to investigate the eukaryotic diversity across the transect. Also included in the data set are control samples including positive and negative controls collected by the autonomous sampler, positive controls samples collected by standard laboratory methods, extraction blanks, and no template controls from PCR amplification.

本数据集依托无人水面艇(Uncrewed Surface Vessel,USV)‘勘测者号’与自动化环境DNA(environmental DNA, eDNA)样本采集设备——环境样本处理器(Environmental Sample Processor, ESP),在北太平洋东部为期29天、航程4200千米的自主航行中,自主采集了环境DNA样本。本研究采用基于Illumina MiSeq平台的细胞色素c氧化酶亚基1(cytochrome c oxidase subunit 1, COI)DNA宏条形码(DNA metabarcoding)测序技术,对该调查样带的真核生物多样性进行了解析。本数据集同时收录了如下对照样本:自主采样器采集的阳性对照与阴性对照样本、标准实验室方法获取的阳性对照样本、提取空白对照以及聚合酶链式反应(Polymerase Chain Reaction, PCR)扩增的无模板对照。
