D. yakuba, D. simulans, and D. sechellia gDNA competitively hybridized against D. melanogaster to evaluate aCGH as a means to identify diverged orthologs. 2 D. sechellia vs D. melanogaster hybs - with dye swap, 2 D. simulans vs D. melanogaster hybs with dye swap, and 8 D. yakuba vs D. melanogaster hybs with balanced dye swaps. D. yakuba vs. D. melanogaster were then analyzed in all 2,4,6,8 possible combinations that incorporated dye-swap to asses sources of variation.
以黑腹果蝇(Drosophila melanogaster, D. melanogaster)为杂交参照,对雅库巴果蝇(Drosophila yakuba, D. yakuba)、拟暗果蝇(Drosophila simulans, D. simulans)与塞氏果蝇(Drosophila sechellia, D. sechellia)的基因组DNA(genomic DNA, gDNA)开展竞争性杂交实验,以评估阵列比较基因组杂交(array Comparative Genomic Hybridization, aCGH)用于识别分化型直向同源基因的可行性。本实验共设置3类杂交组合:2组塞氏果蝇vs黑腹果蝇杂交组(均设置染料交换处理)、2组拟暗果蝇vs黑腹果蝇杂交组(均设置染料交换处理),以及8组雅库巴果蝇vs黑腹果蝇杂交组(采用平衡染料交换设计)。随后针对雅库巴果蝇与黑腹果蝇的杂交样本,采用所有包含染料交换的2重、4重、6重、8重可行组合方案进行分析,以评估变异来源。
