The overall goals and objectives of this study are to investigate the transcriptomics of Neisseria gonorrhoeae using RNA-seq. This work will look at gene expression, start points of transcription, transcriptional termination, and differences between these in different conditions and between strains and growing cultures over time. Overall design: The starting baseline for this study is Neisseria gonorrhoeae strain NCCP11945, minimally passaged since it was genome sequenced, grown at 37C in 5% CO2. All other experiments would be based off of this, both in terms of number of samples and replicates.

本研究的整体目标为采用RNA测序(RNA-seq)探究淋病奈瑟菌(Neisseria gonorrhoeae)的转录组学特征。本研究将聚焦基因表达、转录起始位点、转录终止过程,以及不同培养条件、不同菌株及不同生长时段的培养物间的上述转录相关特征差异。 实验设计概述:本研究的起始基准菌株为淋病奈瑟菌NCCP11945株,该菌株自完成全基因组测序以来仅经过极少传代,于37℃、5% CO₂环境中培养。其余所有实验均以此为参照,样本数量与生物学重复设置均遵循此基准。
