We use a custom microarray for the crustacean Daphnia pulex to investigate gene expression in males, juvenile females and pregnant females. Keywords: sex-biased, developmental Eight biological replicates of 8 day old healthy male Daphnia pulex are compared to four biological replicates of 8 day old healthy (pregnant) females, or to four biological replicates of healthy 6 day old (juvenile) females. Each comparison involved two dye swaps: two males (green) vs. two pregnant females (red), two males (red) vs. two pregnant females (green) and so on.
本研究针对甲壳类动物蚤状溞(Daphnia pulex)采用定制化基因芯片(custom microarray),旨在探究雄性、幼龄雌性与妊娠雌性的基因表达模式。关键词:性别偏倚表达、发育相关。本研究设置两组独立比对:将8日龄健康雄性蚤状溞的8组生物学重复(biological replicates),分别与4组8日龄健康妊娠雌性的生物学重复、4组6日龄健康幼龄雌性的生物学重复进行比对。每组比对均采用两轮染料互换(dye swap)实验设计:具体为2份雄性样本(标记绿色荧光)与2份妊娠雌性样本(标记红色荧光)、2份雄性样本(标记红色荧光)与2份妊娠雌性样本(标记绿色荧光),以此类推。
