Dual-barcoded shotgun expression library sequencing for high-throughput characterization of functional traits in bacteria

Dual Barcoded Shotgun Expression Library Sequencing (Dub-seq) is a method that uses random DNA barcodes to greatly increase experimental throughput. As a demonstration of this approach, we constructed a Dub-seq library with Escherichia coli genomic DNA, performed 155 genome-wide pooled fitness assays in 52 experimental conditions.双条形码鸟枪法表达文库测序（Dual Barcoded Shotgun Expression Library Sequencing，简称Dub-seq）是一种利用随机DNA条形码大幅提升实验通量的技术手段。为验证该方法的实际应用效果，我们构建了携带大肠杆菌（Escherichia coli）基因组DNA的Dub-seq文库，并在52种实验条件下完成了155项全基因组混合适应性测定。
