LiveCyte Migration Raw Data

Live cell imaging and quantitative analysis of cell growth and migration were performed label-free on the Livecyte system (Phasefocus, Sheffield). 3x103 cells (HDLECs and imLECs) in 0.2 ml complete EBM-MV2 medium were seeded per well in 96-well plates (Corning 4580) 4 hours prior to imaging. Before imaging, medium was replaced with complete EBM-MV2 containing 100ug/mL VEGFC or fresh complete EBM-MV2. Cells (four randomly selected 1mm2 field of view per well) were imaged with Olympus PLN 10X (0.25NA) objective for 48 hours at 20min intervals. Single-cell tracking was acquired by Livecyte Analyse software.本研究采用无标记成像方案，于Livecyte系统（Livecyte system，Phasefocus，谢菲尔德）上开展细胞生长与迁移的活细胞成像及定量分析。每孔接种3×10³个HDLECs与imLECs细胞，以0.2 mL完全EBM-MV2培养基重悬后接种于Corning 4580型号96孔板，接种4小时后启动成像。成像前，将孔内原有培养基更换为添加100μg/mL VEGFC的完全EBM-MV2培养基，或更换为新鲜的完全EBM-MV2培养基。每孔随机选取4个1mm²视野，采用奥林巴斯PLN 10×（0.25NA）物镜进行时长48小时的成像，采集间隔为20分钟。单细胞追踪数据通过Livecyte Analyse软件获取。
