Effects of circALTO on gene expression of 293T cells

Purpose: The goal of the study was to determine the potential effects of ALTO expression on the human transcriptome. Method: 293T cells were transfected with circALTO1 or a control plasmid, in triplicate, and mRNA profiles sequenced. Results: mRNA profiles from circALTO1 and control cells were distinct. ALTO expressing cells induced the transcription of multiple genes previoulsy implicated in polyomavirus infections. Overall design: mRNA profile of ALTO1 or vector transfected 293T cells

研究目的：本研究旨在明确ALTO表达对人类转录组（transcriptome）的潜在影响。
实验方法：将293T细胞以三次生物学重复的方式分别转染circALTO1质粒与对照质粒，随后对mRNA（信使RNA）表达谱进行测序。
实验结果：circALTO1转染组与对照组细胞的mRNA表达谱存在显著差异；表达ALTO的细胞可诱导多种此前被证实与多瘤病毒（polyomavirus）感染相关的基因转录。
整体实验设计：对转染circALTO1或空载载体的293T细胞开展mRNA表达谱分析。
