We characterized DNA methylation profiles in blood PBMCs across lentiviral infection in a progressing non-human primate model of AIDS - SIVmac infection in the rhesus macaque. We contrasted methylation data across the course of the SIVmac infection in the young adult and old adult rhesus macaques to identify potential age-related differential methylation patterns and epigenetic clock acceleration. DNA methylation profiles were generated from 40 PBMC samples from 5 old adults and 5 young adults at 4 time points from the baseline across acute infection, early and late chronic infection. For DNA methylation analysis, we utilized custom Infinium methylation array HorvathMammalMethylChip40.
本研究针对以恒河猴SIVmac感染构建的进行性艾滋病非人灵长类模型,解析了慢病毒感染进程中血液外周血单个核细胞(peripheral blood mononuclear cells, PBMCs)的DNA甲基化谱(DNA methylation profiles)。本研究对比了年轻成年与老年成年恒河猴在SIVmac感染全过程中的甲基化数据,以识别潜在的年龄相关差异甲基化模式及表观遗传时钟加速(epigenetic clock acceleration)现象。本次研究的DNA甲基化谱由40例外周血单个核细胞样本生成,这些样本采集自5只老年成年与5只年轻成年恒河猴,涵盖基线、急性感染期、早期慢性感染期及晚期慢性感染期共4个时间节点。DNA甲基化分析采用定制化Infinium甲基化芯片HorvathMammalMethylChip40。
