Using RNA isolated from subcutaneous adipose tissue (SAT) and visceral adipose tissue (VAT) samples obtained from control and class I, II and III obese patients undergoing inguinal hernia repair and laparoscopic cholecystectomy, we compared the gene expression profiles between SAT and VAT using microarrays and validated the findings by real-time quantitative PCR. Two-condition experiment, SAT vs. VAT tissue. Biological replicates: 8 SAT replicates, 8 VAT replicates.
本研究从接受腹股沟疝修补术与腹腔镜胆囊切除术的对照组、I级、II级及III级肥胖患者的皮下脂肪组织(subcutaneous adipose tissue, SAT)和内脏脂肪组织(visceral adipose tissue, VAT)样本中分离RNA,通过微阵列技术比较SAT与VAT的基因表达谱,并采用实时定量PCR(real-time quantitative PCR)对实验结果进行验证。本实验为双组对照试验,比较SAT与VAT组织样本。生物学重复设置为:SAT重复样本8例,VAT重复样本8例。
