Gene expression profiling of 151 pediatric low-grade gliomas

We performed gene expression profiling on 151 paraffin-embedded PLGGs from different locations, ages, histological subtypes as well as BRAF genetic status We also compared molecular differences to normal pediatric brain expression profiles to observe whether those patterns were mirrored in normal brain expression. We analyzed the expression of 6,100 genes among 151 FFPE pediatric and 15 FFPE adult low-grade gliomas and analyzed how the expression patterns changes with location, age, histology and BRAF genomic status and how those differences were mirrored in normal brain expression. 我们对来自不同部位、年龄、组织学亚型以及BRAF遗传状态的151例石蜡包埋的儿童低级别胶质瘤（pediatric low-grade gliomas, PLGG）进行了基因表达谱分析。我们同时将其分子差异与正常儿童脑组织表达谱进行比对，以观察这些表达模式是否在正常脑组织中存在镜像对应关系。我们对151例福尔马林固定石蜡包埋（Formalin-Fixed Paraffin-Embedded, FFPE）儿童低级别胶质瘤及15例FFPE成人低级别胶质瘤的6100个基因的表达情况进行了分析，探究表达模式如何随部位、年龄、组织学类型及BRAF基因组状态发生改变，以及这些差异是否在正常脑组织表达谱中存在镜像对应关系。样本“特征”列的取值含义如下：部位（Location）：SUP=幕上（Supratentorial），INF=幕下（Infratentorial）；组织学类型（Histology）：PA=毛细胞型星形细胞瘤（pilocytic astrocytoma），GG=节细胞胶质瘤（ganglioglioma），DNT=胚胎发育不良性神经上皮肿瘤（dysembryoplastic neuroepithelial tumor），OD=少胶质细胞瘤（oligodendroglial tumors），NOS=未特指肿瘤（not otherwise specified tumors）；BRAF状态：DUP=BRAF基因重复（BRAF duplication），MUT=BRAF V600E突变（BRAF V600E mutation），WT=野生型（wild type），ND=未检测（not determined）；原发或复发肿瘤：P=原发肿瘤（primary），R=复发肿瘤（recurrent）；进展的原发肿瘤：1=是，0=否，_=仅针对复发肿瘤。
