Single-nucleus RNA sequencing reveals that Srsf1 absence alters the cell type composition and differentiation trajectories of BAT. Overall design: Interscapular brown adipose tissue was harvested from 8-week-old control and adipose tissue-specific Srsf1-knockout mice for experiment. In order to obtain single-cell suspension, fresh tissue samples were cut and lysed in nucleus lysis solution. The nuclei obtained were subsequently used for sequencing.

单细胞核RNA测序(single-nucleus RNA sequencing)结果显示,Srsf1缺失会改变棕色脂肪组织(brown adipose tissue, BAT)的细胞类型组成与分化轨迹。实验设计:从8周龄对照组及脂肪组织特异性Srsf1敲除小鼠身上取材肩胛间棕色脂肪组织用于实验。为制备单细胞悬液,将新鲜组织样本切割后置于细胞核裂解液中进行裂解,所获得的细胞核随后用于测序实验。
