To analyze the changes induced in MSC transcriptome by activated T cells, MSC were co-cultured in transwell with anti CD3/CD28 activated T cells for 48 hrs before RNA extraction IFNγ-induced plasticity of mesenchymal stem cells is regulated by STATs through inhibition of mTOR and TGFβ pathways. 为探究活化T细胞对间充质干细胞(Mesenchymal Stem Cells, MSC)转录组的诱导改变,将MSC与经抗CD3/CD28抗体活化的T细胞在Transwell小室中共培养48小时,随后开展RNA提取实验。干扰素γ(Interferon γ, IFNγ)介导的间充质干细胞可塑性,可通过信号转导与转录激活因子(Signal Transducers and Activators of Transcription, STATs)抑制雷帕霉素靶蛋白(mammalian target of rapamycin, mTOR)与转化生长因子β(Transforming Growth Factor β, TGFβ)通路实现调控。该研究的作者包括Vigo T、Procaccini C、Ferrara G、Baranzini S、Oksenberg J、Diaspro A、Kerlero de Rosbo N及Uccella A。本实验将与活化T细胞共培养的MSC(2个生物学重复)的基因表达水平,与未活化的MSC(3个生物学重复)进行了对比分析。
