Spatial Transcriptomic Profiling of the Mouse Adrenal Gland and Adjacent Adipose Tissue

This study employs spatial transcriptomics to map the molecular landscape of the mouse adrenal gland, focusing on distinct cortical zones (zona glomerulosa, zona fasciculata, X-zone) and the adrenal medulla, as well as adjacent adipose tissue. Using Visium CytAssist technology, we analyzed adrenal sections from male CD-1® IGS mice, identifying seven distinct cell clusters, including brown and white adipose tissues. Four sexually mature male CD-1® IGS mice, 10 weeks old with a body weight of approximately 37 g were used in this study. Mice were housed in colony cages in a pathogen-free environment with temperature maintained at 21-23°C and relative humidity between 55-60% and under a 12h light/12 h dark cycle. All mice were given standard chow and water ad libitum. The mice were euthanized by decapitation and the harvested adrenals were immediately preserved in 10% buffered formalin for further analysis.

本研究采用空间转录组学(spatial transcriptomics)技术绘制小鼠肾上腺的分子图谱,重点聚焦肾上腺皮质的三大区域——球状带(zona glomerulosa)、束状带(zona fasciculata)、X带(X-zone)以及肾上腺髓质,同时覆盖邻近的脂肪组织。本研究借助Visium CytAssist(Visium CytAssist)技术,对雄性CD-1® IGS小鼠(CD-1® IGS mice)的肾上腺组织切片开展分析,共鉴定出7个独立的细胞簇,其中涵盖棕色脂肪组织与白色脂肪组织。本研究共纳入4只性成熟雄性CD-1® IGS小鼠,其周龄为10周,体重约37g。小鼠饲养于无特定病原体(Specific Pathogen Free,SPF)环境的群养笼内,环境温度维持在21~23℃,相对湿度为55%~60%,并遵循12小时光照/12小时黑暗的光周期。所有小鼠均可自由采食标准维持饲料并自由饮用饮用水。小鼠经断颈法实施安乐死后,采集的肾上腺组织立即置于10%缓冲福尔马林中保存,以待后续分析。
