We investigated the association between genetic variants in the histone modification regions and the prognosis of lung adenocarcinoma after curative surgery. Potentially functional SNPs were selected using integrated analysis of ChIP-seq and RNA-seq. The SNPs were analyzed in a discovery set (n=166) and a validation set (n=238). The associations of the SNPs with overall survival (OS) and disease-free survival (DFS) were analyzed. This study showed that genetic variants in the histone modification regions could predict the prognosis of lung adenocarcinoma after surgery. 本研究探讨了组蛋白修饰区域遗传变异与根治性手术后肺腺癌患者预后的关联。通过染色质免疫共沉淀测序(Chromatin Immunoprecipitation Sequencing, ChIP-seq)与转录组测序(RNA Sequencing, RNA-seq)的整合分析,本研究筛选出潜在功能性单核苷酸多态性位点(Single Nucleotide Polymorphisms, SNPs)。上述SNPs分别在发现队列(n=166)与验证队列(n=238)中完成分析。随后,本研究考察了这些SNPs与患者总生存期(Overall Survival, OS)及无病生存期(Disease-Free Survival, DFS)的相关性。结果表明,组蛋白修饰区域的遗传变异可有效预测肺腺癌患者术后预后情况。整体实验设计:肺腺癌细胞转录组分析。
