The ventral hippocampus from control and CUMS treated mice was dissected for RNA sequencing. Total RNA was extracted using TRIzol Reagent (Invitrogen, 15596026) according to the protocol provided by the manufacturer . Construction of the cDNA library and sequencing were performed by Guangzhou RiboBio Co., Ltd. (China). Clean reads were obtained after discarding reads containing adapter, poly-N, or low quality raw data. High-quality reads were aligned to the mouse reference genome mm10 with HISAT2. Next, HTseq was used to convert aligned short reads into read counts.

本研究对对照组及慢性不可预知温和应激(Chronic Unpredictable Mild Stress, CUMS)处理小鼠的腹侧海马组织进行解剖,用于RNA测序。按照制造商提供的实验流程,使用TRIzol试剂(Invitrogen,货号15596026)提取总RNA。cDNA文库构建及测序工作由中国广州锐博生物科技有限公司(Guangzhou RiboBio Co., Ltd.)完成。过滤掉含接头序列、poly-N序列及低质量的原始测序读段后,获得洁净读段(clean reads)。使用HISAT2将高质量读段比对至小鼠参考基因组mm10。随后,利用HTseq将比对上的短读段转化为读段计数(read counts)。
