Organs from nonheart-beating donors are attractive for use in cell therapy. Understanding the nature of molecular perturbations following reperfusion/reoxygenation would be highly significant for nonheart-beating donor cells. Nonheart-beating donor rats hepatocytes were studied for global gene expression with Affymetrix microarraays. Groups of rats were used for isolating hepatocytes. The experimental conditions included controls (heart-beating donors) and nonheart-beating rats at various intervals after death as test groups. Each group included samples from three different donor animals.

无心跳供体来源的器官在细胞治疗领域具备显著应用潜力。明确再灌注/复氧后分子扰动的本质特征,对无心跳供体细胞的研究具有极高的科学意义。本研究利用Affymetrix基因芯片(Affymetrix microarray),对无心跳供体大鼠肝细胞的全基因表达情况进行分析。实验通过多组大鼠样本分离肝细胞,设置对照组(心跳供体来源样本)与死亡后不同时间间隔的无心跳大鼠样本作为实验组,每组均包含3头不同供体动物的样本。
