Mouse norovirus (MNV) causes acute or chronic infection in immunocompetent hosts, but the CD8 T cell determinants of viral persistence versus clearance are unknown. We used microarrays to interrogate the transcriptional program on MNV-specific CD8 T cells from the spleen and intestine at days 8, 15, 30 post infection with and acute or chronic strain of MNV. Mice were infected with MNV-CW3 or MNV-CR6fy and tetramer-positive CD8 T cells were sorted from the spleen and intestinal epithelium at day 8, 15, and 30. Analysis was done on an Affymetrix MoGene 2.0 Micorarrays.
小鼠诺如病毒(Mouse Norovirus, MNV)可在免疫健全宿主中引发急性或慢性感染,但目前尚不明确介导病毒持续感染与病毒清除的CD8 T细胞相关决定簇。本研究借助微阵列(microarray)技术,对感染急性或慢性MNV毒株后第8、15、30天的脾脏与肠道来源的MNV特异性CD8 T细胞的转录程序开展检测分析。实验小鼠分别感染MNV-CW3与MNV-CR6fy毒株,并于感染后第8、15、30天,从脾脏及肠上皮组织中分选得到四聚体(tetramer)阳性的CD8 T细胞。本次分析采用Affymetrix MoGene 2.0微阵列芯片完成。
