We infer that non-coding RNAs can function as key effectors during cell cycle, acting on distinct regulatory networks to ensure maintenance of gene expression programs and cell cycle progression. We performed nascent RNA-seq in asynchronized RPE-1 cells as well as synchronized RPE-1 cells at five different timepoints of S-phase of cell cycle progression. We also performed strand-specific RNAseq on control and three knockdown cell lines in which a lincRNA of interest was knocked down. We also performed histone ChIP-seq and Cut-n-run seq on asynchronized RPE1 cells.

本研究推测,非编码RNA(non-coding RNAs)可作为细胞周期进程中的关键效应分子,通过作用于不同调控网络以维持基因表达程序稳定、保障细胞周期顺利推进。本研究对未同步化的RPE-1细胞,以及处于细胞周期S期五个不同时间节点的同步化RPE-1细胞,开展了新生RNA测序(nascent RNA-seq)实验;同时针对对照组及3株敲降目标基因间长链非编码RNA(lincRNA)的细胞系,完成了链特异性RNA测序(strand-specific RNAseq);此外还对未同步化的RPE-1细胞进行了组蛋白染色质免疫共沉淀测序(histone ChIP-seq)与Cut-n-run测序(Cut-n-run seq)实验。
