To further understand the underlying mechanisms and identify genes directly regulated by SETD2 and H3K36me3 on a genome-wide scale, we performed chromatin immunoprecipitation using a H3K36me3 specific antibody followed by next-generation sequencing (ChIP-seq) assays using the renal tubules isolated from SETD2FL/FL KSPCRE/+ and its control SETD2fl/fl mice. 为进一步阐明SETD2与H3K36me3(组蛋白H3赖氨酸36三甲基化)在全基因组尺度上的潜在作用机制,并筛选受二者直接调控的靶基因,我们以从SETD2FL/FL KSPCRE/+小鼠及其对照SETD2fl/fl小鼠中分离得到的肾小管为实验材料,采用H3K36me3特异性抗体开展染色质免疫共沉淀实验,随后进行染色质免疫共沉淀测序(ChIP-seq)检测。此外,本研究还对2种不同的组蛋白修饰在2株原代肾小管上皮细胞中的相关特征进行了分析。
