We performed multiomic high-throughput sequencing to recapitulated the primary mechanism of mesenchymal stem cells osteogenesis. We analysed the H3K27ac ChIP-seq, BRD4 CUT&Tag and RNA-seq data before and after osteogenesis, and identified super enhancers (SEs) regulated osteogenic identity genes in MSCs. 本研究采用多组学高通量测序技术,解析间充质干细胞(mesenchymal stem cells,MSCs)成骨分化的核心机制。我们分析了成骨分化前后的H3K27乙酰化染色质免疫共沉淀测序(H3K27ac ChIP-seq)、BRD4靶向切割与标签化测序(BRD4 CUT&Tag)以及RNA测序(RNA-seq)数据,并鉴定出间充质干细胞中调控成骨特性基因的超级增强子(super enhancers,SEs)。整体实验设计:检测间充质干细胞内BRD4与基因组的结合情况。
