Synovial inflammation contribute to osteoarthritis pathology. Medermycin was isolated from Streptomyces sp. K73 and have antibiotic active against Gram-positive bacteria. Additionally, medermycin exhibits anticancer activity. It has been found that medermycin inhibits the serine/threonine kinase AKT/PKB activity. Akt singnaling modulates NF-kB activation involved in stimulating transcriptional activity of inflammatory processes. We hypothesized that MED may have anti-inflammatory effect via to Akt inhibition in synovial cells. Here, we investigated the effect of medermycin on inflammatory reaction and its mechanism on human synovial fibroblasts.

滑膜炎症(synovial inflammation)参与骨关节炎(osteoarthritis)的病理进程。美达霉素(Medermycin)从链霉菌属(Streptomyces)菌株K73中分离得到,具有抗革兰氏阳性菌(Gram-positive bacteria)的抗菌活性;此外,该化合物还展现出抗癌活性。已有研究表明,美达霉素可抑制丝氨酸/苏氨酸激酶(serine/threonine kinase)AKT/PKB的活性。Akt信号通路可调控核因子κB(NF-κB)的激活,而该激活过程参与介导炎症反应的转录激活环节。我们推测,美达霉素可通过抑制Akt活性在滑膜细胞中发挥抗炎作用。本研究探讨了美达霉素对炎症反应的影响及其在人滑膜成纤维细胞(human synovial fibroblasts)中的作用机制。
